NP-40 buffer is used for mild cell lysis, allowing extraction of cytoplasmic and some membrane-associated proteins while preserving nuclear and chromatin structure.
NP-40 is gentler than RIPA, making it better suited for extracting proteins while preserving interactions. RIPA is harsher and suited for complete cell lysis and membrane solubilization.
